Purpose: Mutations of keratoepithelin KE and myocilin MYOC have been linked to certain types of inherited corneal stromal dystrophy and open-angle glaucoma, respectively. In this study, the feasibility of using small interfering RNAs siRNAs to suppress the expression of keratoepithelin and myocilin and their capabilities to reduce the related cytotoxic effects caused by mutant myocilins were investigated. A BiP promoter-driven luciferase reporter assay was used to evaluate the stress response of TM cells induced by misfolded mutant myocilins. The activation of BiP gene as a stress response induced by mutant myocilins in transformed TM cells was significantly reduced when mutant myocilin proteins were suppressed by myocilin-specific shRNAs. Conclusions: KE- or myocilin-specific shRNAs could effectively suppress the expression of recombinant KE or myocilin proteins and the related cytotoxicity of mutant myocilins.
The development of a complex nervous system requires the actions of intricate genetic mechanisms that influence and maintain the differentiation of common nervous system progenitor cells into neurons and glial cells. In Drosophila melanogaster, the gene glial cells missing gcm encodes the transcription factor Gcm that, when active, causes early nervous system cells to preferentially differentiate into glial cells by activating the reversed polarity repo gene, as well as others. However, the Gcm protein also plays a role in the differentiation of both macrophages and tendon cells.

It is possible to find usable subsequence on its pursuant biochemical properties and structural characteristics. Specifically, this work studies relationship between mentioned theoretically calculated property of subsequences and their efficiency for RNAi for any input set of experimental data. Furthermore, in this work software RNA Interference Analyzer is designed, implemented, tested and used.
